Brazilian striker Yan Sasse scored the winning goal to help Tunisian side Esperance beat Mamelodi Sundowns 1-0 in Tunis on Saturday night.
The goal arrived in the 41st minute with Sasse round off goalkeeper Ronwen Williams after a clever ball from Rodrigues.
Sundowns created several good opportunities to equalise in the second half but were not clinical.
They will have to overturn the deficit in a week’s time in Pretoria.
In Lubumbashi, holders and record champions Al Ahly frustrated hosts TP Mazembe as the two sides fired blanks.
The match will be decided at Cairo International stadium next week.
